Malarone (Atovaquone and Proguanil)
Purpose of Medication and Method of Action
Malarone is indicated for both the prevention and treatment of malaria. It combines two drugs. Atovaquone which interferes with mitochondrial transport (and energy production) of the malarial parasite while proguanil inhibits a parasite enzyme required for normal protein synthesis, dihydrofolate reductase.
Side Effects
The most common side effects include: abdominal pain (17%), nausea (12%), vomiting (12%) headache (10%), diarrhea (8%) and weakness (8%).
Drug Interactions
Concomitant treatment with tetracycline, metoclopramide and rifampin will reduce blood levels of atovaquone and tetracycline should be avoided if possible.

Usual Dose
For malarial prevention, one tablet at same time each day with a meal or milk. If vomiting occurs within hour, repeat dose. Acute malaria - four tablets as a single dose for 3 consecutive days.
